I to am having the same issue. I opened the game and tried to click and thing and my mouse does icon does not align up with where it see my mouse. to Select a character my mouse is all the way to the left of my screen. I canNOT go any further. I followed the so call steps with closing other PROGRAMS! Yea that didn’t help.

While you are in game go to options and then down to the mouse/keyboard tab. You will want to look for the option Confine Mouse Cursor. I recommend setting it to Smart or simply leave it On. This should make sure your mouse stays confined to the window.
Also make sure that the Windows desktop resolution option matches the in-game resolution. Then check that the Scale and Layout option in Windows is set to 100%.
here is the issue with that. I canNOT click on options because my mouse even tho the icon shows it is over the options. IS NOT seen as being there. I can’t even close without using the “End Task” in the task manager.
I cannot play HotS due to the fact my mouse pointer is not where the game sees it. I have followed other stuff I found in the forums about shutting down startup programs (which honestly should not be needed to be done at all). I’ve shut down all but 4 which are needed to run the computer but still it’s way off.
BLIZZARD needs to fix their stuff! Why I spot playing blizzard games a long time ago. I hoped after a few years they would of fix these issues.

Typically when I see the pointer in a wrong location, it is related to either Resolution or Scaling.
Lets try opening up the Display Settings in Windows. In this menu ensure “Change the size of text, apps, and other items” is set to 100% (Recommended). We’ll also want to adjust the Resolution setting to any (Recommended) value if it is not already.
We’ll also want to open up the in-game settings menu, and ensure the Resolution matches the Recommended value for Windows.
Yeah, i have the same issue after installing the game again.
My windows display settings are set to 100%.
it’s like the resolution is twice as big and then reduced to fit my resolution, but the buttons are like fixed at a certain pixel, which does not match the pixel of the button or something.
I also tried to reduce the screen resolution by using my nvidia experience settings. but so far the same result every time.
Also i cannot find a .config file with the setting of the game so i can change them there.
I think it has to do with the game setting it automatically to 3840x2160 DSR but the pointers don’t scale along with that for some reason.
ok, i found a temp fix.
go into blizzard settings, then game setting, the scroll down to HOTS and reset in game options. after that i was able to enter ingame settings.
