The troll is Vol’jin.
The robot with the hat is Bob, the best! The little animal in his pocket is Hammond the Hamster that is Wrecking Ball. The white-haired cowgirl is Ashe. Ashe & Bob are a Hero together.
Red-eyed fella is Wrathion.

As a guess, Diablo Druid (middle) and Baal, Lord of Destruction (right).

Baal is really the odd one out as far as HotS goes. Both Diablo and Mephisto are in HotS but Baal, the literal title sake of the Diablo II expansion, is still missing.

I would really like to see Baal be added to HotS, even if it is just a special out the way once off thing done to tie in and promote Diablo IV.

Btw, I’m a fan of the netcomics Cyanide & Happiness, they have depressing comic weeks (since they post daily) and I liked the idea.
I also like to make thematic comics and planned releases.
