Alright, I know this is an old meme and I am well aware of Kael’thas saying it. However, I don’t know what about this makes the meme a funny one, or why it even became a meme in the first place?
Because it was funny that he was acknowledging that we already murdered him once, and he was super extra about it while doing so.
Also it’s a meme. You can’t explain memes. They just happen.

It’s a running joke that started with Kael, but over the years other characters say it too and reinforced it. It’s poking fun at the fact that some enemies just don’t die the first time.
Ragnaros, Nefarion, Anub’arak, Kael’thas, most troll enemies, and many many others too. The first loss was just merely a setback until they come back and try again later on.

It’s a meme because, for one, it’s a speech most people will remember hearing; and second, the phrase “merely a setback” can be applied to many other contexts, so the meme potential is high.
