Does Envenom do less damage if the target

…doesn’t have rupture and garrote on them?

Already have enough dots out for the energy regen so it’s time to envenom. Should you envenom a target with both dots or does it not matter?

Envenom is not affected by DoTs.

Envenom does not do more or less damage with Dots on the target, however you should never let Garrote or Rupture fall of the target, if Rupture is close to falling off you will instead use those Combo Points to refresh it and not use it on Envenom.
