Impale doesn’t work properly when using a range weapon. Now I know people will say that its normal cause you need a dagger equip but that’s also the point. You need a range weapon to use some skills which is fair but you don’t need a melee weapon to equip Impale so it’s my understanding that its by design?
This is no useful error description. What does not work?
When Karlei’s Point is equip the buff work fine but when it’s in the Kanai’s Cube and you equip a range weapon the Karlei’s Point buff don’t apply.
Are you sure it’s not the 2 pc Shadow set bonus you’re losing (you need a melee weapon for that)? Did you test without the Shadow set altogether?

I should specify that that’s also considering that the 6000% buff when having a melee weapon equipped of the Shadow set is not active. (Unless im wrong)
