I leveled all my current build glyphs past 45 except IRE and Might. It doesn’t give me the option to upgrade. Ran pit multiple times, removed glyph, added glyph, power cycled, ran in a circle and patted my head. Still nothing. Bummer please hot fix this blizzard 
It costs gem fragments to upgrade to 46. Do you have enough?
